A short protocal version
membrane → booster → blocking → antibody(sample) → ECL
Important notes:
- Signal enhancement is better for NC than PVDF membrane.
- Signal enhancement is better for PVDF that is stored in water or PBS.Dried PVDF membrane is more resistant to western blot booster.
Procedure for Western blot booster
- Wash the membrane (NC or PVDF) with deionized water three times, each for few seconds.
- Cover the membrane with solution A and allow for incubation for 5 min at room temperature.
- Wash the membrane with deionized water three times, each for few seconds.
- Cover the membrane with solution B and allow for incubation for 5 min at room temperature.
- Proceed with blocking the membrane with skim milk or other appropriate blockers.This treatment of membrane would lead to enhanced signal intensity by several folds.
Storage and Stability:
Stable when stored at room temperature. Use sterile condition for removing solution from the container, when used properly it is good for one year to use.
